TypicaltransformationresultsThefollowingtransformationswereperformedatthesametimeusingthemultiwelltransformationprotocol.SimilaramountsofPCRproductwereusedforeach.ThepicturesdemonstratethedegreeofsuccessandvariABIlitythatcanbeattained.Theplateswereincubatedfor6daysat30°Cbeforebeingphotographed.Thehaploidplateshowsthemanysmallerbackgroundcolonies(1mm)whichappearearlyduringtheincubation,inadditiontothetruetransformants(3-10mm).ThesesmallercolonieswillnotshowresistancetoG418whenrestreakedanddonotappearinthediploidstrain.Colonysizedependstosomeextentonthenumberofpositivesontheplate,ontheamountofbackgroundandonwhetherthedipruptedgenewasimportantforviability.Thetransformationefficiencyofthediploidstrainisapproximatelyone-thirdofthehaploidstrainforunknownreasons.(OnepossIBLityisthatfewerdiploidcellsareusedinthetransformationperOD600thanthehaploid.Thisneedstobetested.)
