- Cut1/2"-3/4"ofmousetail.(Checkwithyourinstitution"sAnimalStudiesCommitteefortheirrecommendationsastohowthisprocedureshouldbeimplemented).
- Addtailfragment(ortissueofthesamesize)directlyto500ulofextractionbufferandadd50ulofProteinaseK.
- Incubateovernightat52o-55oC.
- Add500ulofTrispH8.0equilibratedphenol.Mixvigorously,spin(12,000-14,000RPM)atroomtemperaturefor2minutes.
- Removeaqueousphasetoafreshtube.
- TotheaqueousphaseaddanequalvolumeofChloroform/IsoamylAlcohol(49:1).Mixvigorously.Spinatroomtemperaturefor2minutes.Removeaqueousphasetoafreshtube.
- Totheaqueousphaseadd250ul7.5MNH4OAcand750ulisopropanol.Mixwell.Spinatroomtemperaturefor5minutes.Washthepelletoncewith70%ethanol.
- Drythepelletusingaspeedvac.
- ResuspendtheDNAin400ul1XTE;add2ulRNAseA(10mg/ml)andincubateovernightat37oC.
- Storeat4oC.Cut100ul(approximately10ug).
