AcrylamideSolutions
AcrylamidesolutionsforPAGEaregivenastotalconcentrationofacrylamide(acrylamide+bisacrylamide)andtheamountofcrosslinker(bisacrylamide).ThisislistedastheT:Cratio.Forexample,a10%gel(10%T:5%C)wouldcontainatotalof10gramsofacrylamideper100ml,andwouldbecomposedof5gramsofacrylamideand5gramsofbisacrylamide.Usually,astocksolutionof30%acrylamideisproducedcontaining0.8%bis-acrylamide.Manyinvestigatorsuse30gramsofacrylamideplus0.8gramsofbis-acrylamideper100mlofwater,but29.2gramsofacrylamideplus0.8gramsofbiswouldbetechnicallycorrect.Inpractice,itmakeslittledifferencesincethegelsaredilutedto10%orless.The30%stocksolutionisfilteredthrougha0.45µfilterandstoredat4°Cinthedark.Foruse,thestocksolutionisdilutedwithanappropriatebuffer(usuallya2XTris-HCl).Thestocksolutionisstableforaboutonemonth.Discardafterthisperiod.
