Toreplicateinthehostcell,virusescommandeercellularsignalingpathways.Cytomegalovirus(CMV)isaDNAviruswiththatiswidespreadinthepopulationbutusuallycausesdiseaseonlyinimmunocompromisedindividualsandisalsoaviralcauseofbirthdefects.OneoftheactionsofCMVinthehostcellistostimulateMAPkinasepathways.Bothp38andERKkinasesareactivatedbyCMVinfectionthroughactivationmapkinasekinasesandinhibitionofphosphatases.OneresultofMapkinaseactivationbyCMVisactivationoftranscriptionofviralgenes,increasingtheproductionofviralgeneproducts.Bothp38andERKkinasescontributetotheactivationofviralgenesbycellulartranscriptionfactorsactingthroughtheviralUL4promoteratupstreamandbasaltranscriptionelements.Anothertargetofprolongedp38activationduringinfectionisRb,contributingtoviralreplication.ActivationofMKK1andMKK2leadstoErk1andErk2activation,andphosphorylationofdownstreamtargets.TheMEKK1kinaseregulatestheimmediateearlypromoterindirectlythroughdownstreamkinasesignalingandperhapsmoredirectlythroughactivationofNF-kB.MapkinasepathwaysactivatedbyCMVconvergeonincreasedtranscriptionofviralgenesandincreasedreplicationoftheviralgenome.BetterunderstandingofthemechanismsinvolvedintheinteractionofCMVwithcellularsignalingmachinerywillprovideimprovedwaystotreatCMV-mediateddisease.
