- Decitabine is a cytosine nucleoside analog and inhibits further DNA methylation after incorporated into DNA. As a consequence, tumor suppressor genes aberrantly silenced after methylation can be reactivated and expressed. Decitabine has shown activity in some hematologic disorders including myelodysplastic syndromes (MDS), acute myelogenous leukemia (AML) and chronic myelogenous leukemia (CML). - Treatment of patients with sickle cell disease (SSD) with subcutaneous decitabine increased fetal hemoglobin (HbF) levels and improved SSD pathophysiology such as red cell adhesion, endothelial damage, and coagulation pathway activity without cytotoxicity. Saunthararajah Y., et al. \"Effects of 5-aza-2-deoxycytidine on fetal hemoglobin levels, red cell adhesion, and hematopoietic differentiation in patients with sickle cell disease.\" Blood 102: 3865-3870 (2003).
