Aquaporin 2 phospho S261 Antibody Description

Background Aquaporin 2 pS261 antibody recognizes aquaporin 2 (AQP2) which is a hormonally regulated water channel located in the renal collecting duct. Mutations in the AQP2 gene cause hereditary nephrogenic diabetes insipidus in humans. A vasopressin induced cAMP increase results in the phosphorylation of AQP2 at serine-256 and its translocation from the intracellular vesicles to the apical membrane of principal cells. Recently, serine-261 has been identified as a novel phosphorylation site on AQP2 and levels of phosphorylated S261 have been shown to decrease with vasopressin treatment suggesting its involvement in vasopressin-dependent AQP2 trafficking. Application Note Anti-Aquaporin 2 pS261 antibody is suitable for use in ELISA, Western Blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band of approximately 29 kDa AQP2 protein phosphorylated at Ser261 in the appropriate cell lysate or extract. This antibody also recognizes the glycosylated for of aquaporin 2 at approximately 37 kDa in size.

Purity/Specifity Anti-Aquaporin 2 pS261 Antibody is directed against rat Aquaporin 2 protein phosphorylated at S261. The antibody was prepared from monospecific antiserum by immunoaffinity chromatography using phospho peptide coupled to agarose beads followed by solid phase adsorption(s) against non-phospho peptide and non-specific peptide to remove any unwanted reactivities. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum. Minimal reactivity occurs against non-phosphorylated Aquaporin 2. Reactivity against Aquaporin 2 occurs from rat sources. However, reactivity is also expected against the following species based on 100% sequence homology: canine and bovine, chicken, human, mouse and non-human primate.
