Background

AKING1 is a member of SnRK1/SNF1/AMPK family in plants. It is a regulatory subunit of the putative trimeric SNF1-related protein kinase (SnRK) comples which may play a role in a signal transduction cascade regulating gene expression and carbohydrate metabolism in higher plants.

10 ug of Arabidopsis thaliana Columbia WT and KIN gamma KO seedlings were grinded in MOPS loading buffer and heated for 5 min at 95°C. Samples were separated on 10 % SDS-PAGE and blotted 1h to PVDF. Blots were blocked with 5% milk for 1h at room temperature (RT) with agitation. Blot was incubated in the primary antibody at a dilution of 1: 2 000 for 2h at RT with agitation. The antibody solution was decanted and the blot was rinsed briefly twice, then washed once for 15 min and 3 times for 5 min in TBS-T at RT with agitation. Blot was incubated in secondary antibody, diluted to 1:4 000 in for 1h at RT with agitation. The blot was washed as above and developed for 5 min with ECL according to the manufacturers instructions. Exposure time was 15 seconds.
