DescriptionMouseCTLA-4wasinitiallyidentifiedbydifferentialscreeningofamurinecytoliticCDNAlibrary.ItisamemberoftheIgGsuperfamily,anditencodesa223aminoacidproteinandpossessesasingleextracellularVdomain,atransmembranedomain,andacytoplasmicdomain.CTLA-4is~30%homologouswithCD28,andbothbindtothesameligands(B7.1/CD80andB7.2/CD86)althoughCTLA-4bindswithhigheraffinitythanCD28.ParallelrecognitionofaspecificMHC-peptidecomplexbytheT-cellreceptorandCD80orCD86bythecostimulatoryreceptorCD28resultsinT-cellactivation,cytokineproduction,proliferation,anddifferentiation.AftertheTcells"activationprocessandupregulationofCTLA-4,co-ligationoftheTCRtoMHC-peptidecomplexandCTLA-4toitsligandsresultsincellcyclearrestandtheendTcellactivation.IthasbeensuggestedthatthenegativeregulatorymechanismofCTLA-4ismediatedthroughtrans-endocytosisofCD80andCD86(captureofligandsfromopposingcells).TheligandsareremovedfromtheoriginalcellanddegradedinsideofCTLA-4-expressingcells.Asaresult,theactionofCD28isimpaired.AlternativesplicevariantsofCTLA-4havebeendescribedinmiceandhumans;thesevariantsskipexon2(ligand-bindingdomain)orexon3(transmembranedomain).TheligandindependentCTLA-4isexpressedinmice,butnotinhumans,andthesolubleCTLA-4(sCTLA-4)isexpressedinmiceandhumans.CTLA-4isconstitutivelyexpressedbyTregs,andtheyareanimportantsourceofsCTLA-4.IthasbeensuggestedthatsCTLA-4potentiatesregulatoryTcellfunction.HighlevelsofsCTLA-4inTregshavebeenassociatedwithtype1diabetes(T1D)andotherautoimmunediseases.ProductDetails
