Description: IL-12 is a potent regulator of cell mediated immune responses and it induces IFN-g
production by NK and T cells. It is produced by activated monocytes/macrophage cells, B lymphocytes
and connective tissue type mast cells. Among its biological activities IL-12 promotes the growth and
activity of activated NK, CD4+ and CD8+ cells and induces the development of IFN-g producing Th1
cells. Recombinant murine IL-12 is a 75.0 kDa heterodimeric glycoprotein consisting of disulfidelinked
35 kDa (p35) and 40 kDa (p40) subunits (506 total amino acid residues).

Endotoxin level: Endotoxin level is less than 0.1 ng per mg (1EU/mg).
Formulation: Sterile filtered through a 0.2 micron filter. Lyophilized from 0.5x PBS, pH 7.2.
Reconstitution: Centrifuge the vial prior to opening. Reconstitute in water to a concentration of
0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and
stored at 4oC for 1 week or -20oC for future use.
Stability: The lyophilized protein is stable for a few weeks at room temperature, but best
stored at -20oC. Reconstituted IL-12 should be stored in working aliquots at -
20oC.
Biological Activity: The ED50 was determined by the stimulation of IFN-g production by murine
splenocytes co-stimulated with IL-12 is < 0.1 ng/ml, corresponding to a specific
activity of > 1 x 107 units/mg.
