Short Description: In this assay the HEMOPEXIN present in samples reacts with the anti-HEMOPEXIN antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-HX antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound HX. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3’,5,5’-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of HX in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of HX in the test sample. The quantity of HX in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Kit Principle Genway Biotech Hemopexin ELISA Kit (Human) (GWB-4B6D1A) is based on standard sandwich enzyme-linked immuno-sorbent assay technology. An antibody for Hemopexin has been pre-coated onto a 96-wellplate (12 x 8 Well Strips). Standards or test samples are added to the wells, incubated and washed to remove unbound proteins. An Anti-Hemopexin - HRP conjugated detector antibody is then added, incubated and unbound conjugate is washed away. An enzymatic reaction is produced through the addition of TMB substrate which is catalyzed by HRP generating a blue color product that changes to yellow after adding acidic stop solution. The density of yellow coloration read by absorbance at 450 nm is quantitatively proportional to the amount of sample Hemopexin captured in the well. |
