application.Enzymicproceduresvaryinpre-treatmentsteps,2starch
gelatinisation,liquefactionanddextrinisation,hydrolysisofdextrinsto
GlucoseandGlucosemeasurement3.AACCMethod76-114specifies
starchgelatinisationunderaqueousconditionsinanautoclave,
followedbystarchconversiontoGlucosewithamyloglucosidaseand
Glucosemeasurement.AACCMethod76-11underestimatesstarch
contentinarangeofsamplesandmaterials,includinghighamylose
maizestarchesandmanyprocessedcerealproducts.Mostmethodsin
usetodayincorporatetreatmentwithThermostableα-amylaseeither
duringorimmediatelyfollowingthestarchgelatinisationstep.5,6For
sampleswhicharedifficulttogelatinise(suchashighamylosemaize
starch)solventssuchassodiumhydroxideordimethylsulphoxide
(DMSO)7,8havebeenemployed.Inaproceduretoensurecomplete
solubilisationofstarchindietaryfibredetermination,Englystand
Cummings(1988)7includedtreatmentwiththestarchdebranching
enzyme,pullulanase.
Tosatisfytheneedforanextremelysimple,yetquantitativeand
reliable,procedureforthemeasurementoftotalstarchMegazyme
produces,andoffers,atotalstarchassaykitbasedontheuseof
thermostableα-amylaseandamyloglucosidase(McClearyetal.9).This
methodhasbeenadoptedbyAOAC(OfficialMethod996.11)and
AACC(Method76.13).
Morerecently,thermostableα-amylasesthatareactiveandstable
atlowerpHvalueshavebecomeavailable.Consequently,wehave
updatedourtotalstarchmethodologytoincorporatesuchan
enzyme10.Themajoradvantageofthisimprovementistoallowboth
thethermostableα-amylaseandamyloglucosidaseincubationsteps
tobeperformedatthesamepH(pH5.0)which,inturn,simplifies
theassayandminimisesthepossibilityofproductionofmaltulose
(4-α-glucopyranosyl-D-fructose)11,whichisresistanttohydrolysisby
amyloglucosidaseandα-amylase.
TheMegazymetotalstarchanalysisprocedureallowsthe
measurementoftotalstarchinawiderangeoffood,feed,plantand
cerealproducts(naturalorprocessed).Formostsamples(e.g.wheat
flour),starchiscompletelysolubilizedonincubatingthesampleat
approx.100°Cinthepresenceofthermostableα-amylase.Samples
containinghighlevelsofresistantstarch(e.g.highamylosemaize
starch),requirepre-dissolutionincold2MKOHorhotDMSO.For
samplescontainingsolublestarchormaltodextrins,cookingwith
thermostableα-amylaseisnotrequired.
