花菁类染料-美国Lumiprobe

花菁这类染料的分子内部含有由甲川基(CH)n组成的共轭链,n可为奇数或偶数。共轭链两端或链中间连有杂环、芳环化合物、环烯化合物等与共轭链组成一个大的共轭体系,分子内部的氢可被一定数目的各类取代基取代。

基于其结构,花菁通常有非常高的超过100000 Lmol-1cm-1消光系数。不同的取代基具有控制发色团的性能,如吸收波长、耐光性、荧光。例如,通过聚次甲基桥键长度的选择来控制吸光度和荧光波长:聚次甲基桥键越长其吸光度越高且发射波长可达到近红外区。

已经有许多的花菁染料(Cyanine dyes)用于生命科学领域,一系列的噻唑和恶唑染料用作DNA 和蛋白质染色(如TOTO,YOYO, Stains All和其它)。但是在生命科学研究领域最受欢迎的花菁染料是由卡内基梅隆大学的Alan Waggoner和他的同事们在1990年代早期发现的。该花菁染料是经过修饰后的吲哚菁绿(ICG),从1970年代以来该染料一直用于血管造影术,而 且他们都包含两个假吲哚环侧翼(CH)n链。发现此类染料对生物分子表现出较低的非特异性结合, 由于巨大的消光系数能发出明亮的荧光和良好的量子产量。一旦获得专利, 在卡内基梅隆专利过期后这些分子可在公共领域获得,并从Lumiprobe可购买到, 作为各种反应衍生物用于科研和商业,比如用于点击化学的NHS酯、马来酰亚胺、叠氮化物的衍生品和其他衍生物。

花菁染料分为两种:非磺化的花菁染料和磺化的花菁染料,对于许多的应用,他们是可以互相替换的,因为他们的光谱属性几乎 相同。磺化和非磺化染料可用于DNA和蛋白质等生物分子的标记。两种染料的区别在于它们的溶解度:磺基染料是水溶性,他们可在水环境中标记无需添加有机助 溶剂,他们在水里的分散性较好。在某些情况下,你可能会只需要这两种染料中的一种(下面将具体讲述非磺化的花菁染料和磺化的花菁染料的具体区别。)

非磺化的花菁染料

非磺化的花菁染料的荧光性质对溶剂和反应环境的依赖性较小,非磺化的的花菁染料的吸光度和荧光光谱绘制如下。

磺化的花菁染料

磺化的花菁染料包含了额外地磺基官能团能促进染料分子在水相中的溶解度,带电荷的磺酸盐降低了染料分子的聚合度且能很好地标记偶联物。

目前可用的磺化花菁染料包括sulfo-Cy3,sulfo-Cy5和sulfo-Cy7。

磺化花菁染料具有高度水溶性。标记生物分子时不需要有机助溶剂辅助。

磺化和非磺化花菁染料之间的异同

磺化和非磺化花菁染料都展现出非常相似的荧光性能。然而,在使用说明上有几个不同点需要注意,非磺化花菁染料必须要先溶 解在有机助溶剂中(DMF or DMSO),之后在适当的水溶缓冲剂作用下加入靶标生物分子溶液。推荐使用的助溶剂的体积为,溶解Cy3, Cy5, Cy7使用10%助溶剂,.5的染料使用15%的助溶剂。而磺化的花菁染料可以在纯粹的水环境中应用。关于花菁染料在纯化方面的差异:当使用与水或水溶缓 冲溶液相关的透析纯化染料时,必须使用sulfo-Cy以实现高效除去未反应的染料材料。与磺化和非磺化花菁染料的反应可以通过凝胶过滤,色谱法 (FPLC,高效液相色谱法,离子交换法),或电泳纯化。

磺化和非磺化花菁染料在标记以下类别目标时是可互换的:

1.可溶性蛋白,其对额外添加的有机助溶剂具有耐受性

2.抗体(使用5 10% DMSO / DMF)

3.DNA和寡核苷酸

4.肽

5.许多小分子

This high load, 500 controlled pore glass solid support is designed for the synthesis of oligonucleotides containing up to 50 bases with a 3 -amino group. Amino group is protected with trifluoroacetyl (TFA) protection which is easily removed under standard deblock conditions.

Amino-11-dUTP is an improved replacement for Aminoallyl dUTP. This triphosphate features longer linker, which allows for easier and more facile downstream labeling.

Amino-11-dUTP is compatible with PCR, nick translation, and reverse transcription. It can be used for microarray labeling. Pyrene is polycyclic aromatic hydrocarbon which is well-known for its ability to intercalate into DNA. Pyrene features intense blue fluorescence. Its parameters depend strongly on fluorophore microenvironment. Therefore, fluorescence spectra of pyrene are used for the extraction of structural information about site surrounding pyrene. Two pyrenes in close proximity usually form excimers easily detectable by excimer fluorescence. Pyrene can also be a FRET donor to other fluorophores such as perylene.With this phosphoramidite, pyrene can be introduced into DNA or RNA by means of automated synthesis. This pyrene phosphoramidite contains hydrocarbon moiety rigidly attached to deoxyuridine. This reagent allows to attach pyrene fragment to 5 , internal, or 3 position (using universal support).This amidite requires no special handling, coupling, or deprotection conditions. Recommended diluent for it is acetonitrile.

This ROX formulation is a passive reference dye for qPCR application. The use of this dye is recommended for the normalization of fluorescence intensity of qPCR reporter dyes, such as SYBR Green I or TaqMan probes. Long trebler phosphoramidite is a branching reagent for oligonucleotide synthesis allowing to synthesize branched DNA structures using standard DNA synthesizer.After the condensation of the trebler, three DNA branches begin to grow simultaneously with each step of the synthesis. Deblock of this construct gives rise to DNA containing branching point. One arm (stem) is attached to branch point with its 5 -end, and other arms (branches) are attached via 3 -end. Reverse amidites can be used to prepare constructs with different branch orientations. Repetitive condensations of trebler results in formation of DNA dendrimers.Trebler amidite can be used to attach several modifier amidites to 5 -end of oligonucleotide for example, three biotin residues can be attached at once.This amidite does not require any special handling. Recommended coupling time is 5 minutes. No changes to deblock conditions are required.

DMF (dimethylformamide) is a recommended solvent for labeling reactions using NHS esters. However, when DMF is stored in air, it deteriorates slowly absorbing moisture, and releasing dimethylamine. These impurities rapidly destroy NHS ester function, decrease labeling efficiency, and even make labeling reaction fail.

Dimethylformamide solvent requires special drying and preparation, which is often not available in all laboratories.

This product contains ready-to use solvent which is guaranteed to be useable for the labeling. Tubes with high quality, dry, and amine-free DMF are packed under argon.

A carbonyl-reactive hydrazide derivative of Cyanine7.5 near infrared (NIR) fluorophore. This dye has similar spectral properties with ICG (indocyanine green), but has a higher quantum yield of fluorescence. Hence, this fluorophore is ideal for in vivo imaging.

This is a carbonyl-reactive dye which is particularly useful for the labeling of antibodies after periodate oxidation. It also labels other carbonyl compounds (aldehydes and ketones), for example, many saccharides.
