SMARTer Stranded Total RNA-Seq Kit - Pico Input Mammlian은 Picogram 수준의 total RNA (250 pg - 10ng)로부터 Illumina sequencing library를 효율적으로 제작할 수 있다.또한 high-quality, partially degraded RNA 뿐만 아니라 low quality total RNA (FFPE 또는 LCM 샘플등)로부터도 NGS library 제작이 가능하다. 본 제품은 strand information을 정확하게 유지할 수있을 뿐 아니라 cDNA 합성 이후 별도의 librarypreparation 과정 없이 역전사과정과 PCR 증폭과정에서 Illumina sequencing 분석용 index와 adaptor를 부착할 수 있다. Ribosomal RNA (rRNA)는 total RNA의 약 90%를 차지하는 RNA로써 NGSlibrary 제작 전, rRNA를 제거(depletion)함으로써제작비용을 절감할 수 있을 뿐만 아니라 mapping statistics 또한 향상시킬 수 있다. 하지만 매우 극소량의 Total RNA를 이용할 경우 일반적인 방법의 rRNA depletion은 NGS library 제조하는 데 필요한양에 미치지 못 하게 된다. 본 제품에 사용된 기술은 mammalianrRNA와 mitochondrial RNA 특이적인Probe를 이용하여 rRNA derived cDNA를 제거한다. High-troughput version인 SMARTer StrandedTotal RNA-Seq Kit - Pico Input Mammalian (Code 635007)은96개의 index library 제작을 위한 12개의 reverse primer와 8개의 forward primer를 포함하고 있다.
