The3G3antibodyreactswithmouseFoxp3,a50-55kDatranscriptionfactorwhichisacentralregulatorofTcellactivityandiscriticalforthedevelopmentandfunctionofregulatoryTcells(Tregs).Foxp3isexpressedatconstitutivelyhighlevelsinTregcells,whicharefurtheridentifiedasbeingCD4+CD25+.InrestingconventionalTcells(CD4+CD25-)Foxp3expressionisrestricted,anduponTCRactivationisexpressedonlytransientlyandinasmallproportionofcells.However,thegrowthfactorTGF-betahasbeenshowntoinduceexpressionofFoxp3innaïveTcells,drivingtheirdevelopmentintoFoxp3+Tregs,whicharecalled“induced”or“adaptive”Tregs.Thesecellsarephenotypicallysimilartoso-called“natural”Tregs(CD4+CD25highFoxp3+)whichoriginateinthethymusandcomprisethemajorityofTregcells.Tregsarecriticalformaintainingperipheraltoleranceandareimplicatedinthedevelopmentofautoimmunity.
ItisimportanttoreviewtheliteratureinchoosinganantibodyfortheFoxp3antigeninflowcytometry,asthepotentialforhighbackgroundornon-specificstainingmaybeobserved.The3G3antibodymaybeusedforintracellulardetectionofFoxp3incellsfrommouseandRhesusmacaque.
