InVitro

JC-1(2.5 μM)exposedtomurineL1210lymphoblasts,canbedetectedthepresenceofbothcytoplasmicJC-1monomerandmitochondrialJ-aggregatesinthesecells.JC-1fluorescenceisusuallyexcitedbythe488 nmlaserwavelengthcommoninflowcytometers[1].FluorescentlabelingofmitochondriawitheitherJC-1(1μg/mL,15min),revealsthataredistributedirregularly,resultinginregionsofhighandlowmitochondrialcontentwithinastrocytes[2].JC-1hasbeenshowntointeractwithα-synucleinattheacidicC-terminalregionwithaKdof2.6μM.JC-1itselfdoesnotacceleratetheproteinaggregationofα-synucleinintheabsenceofiron,insted,itdeceleratestheaggregationprocessbyextendingthelagphaseapprox[3].JC-1isavidlyaccumulatedinsensitiveK562cellswhereitdisplaysbothagreencytoplasmicandredmitochondrialfluorescence.JC-1ispoorlyaccumulatedinresistantK562cells,whichdisplaysonlyaslightgreenfluorescence[4].
