MitomycinCisproducedbyastrainofactinomyces, Streptomycescaespitosus.Itcontainsthreeanticancermoieties,quinine,urethane,andaziridinegroups.Itisusedtogeneratemitoticallyinactivefeedercellsincellculturesystems,suchasthemitoticallyinactivefibroblastsusedinembryonicstemcellsystems. Thisproductisan alkylatingagent thatspecificallytargetstheguaninenucleosidesequence5′-CpG-3′.ItinhibitsDNAsynthesisbycovalentlyreactingwithDNA,formingcrosslinksbetweencomplementarystrandsofDNA.ThisinteractionpreventsseparationofcomplementaryDNAstrands,inhibitingDNAreplication.MitomycinChasstrong antitumoractivity,especiallyagainstEhrlichascitestumorcells,andstrongbactericidalactionagainstgram-positiveandgram-negativebacteria.
