The reaction was done in 50 mM Tris-HCl, pH 7.5, 12.5 mM MgCl2, 1 mM β-D-fructose-6-phosphate, 100 μM ATP, 2 mM DTT, 5 mM KH2PO4, and various amount of PFKFB3 in a final volume of 50 μL. The reaction was incubated at 30°C for 30 min, and the amount of ATP consumed was calculated using Kinase-Glo® Plus reagent (Promega).
