DescriptionTheImmunoStarN-terminalneuronalnitricoxidesynthaseantiserumwasqualitycontroltestedusingstandardimmunohistochemicalmethods.Theantiserumdemonstratesstronglypositivelabelingofrathypothalamus,striatum,cortexandspinalcordusingindirectimmunofluorescentandbiotin/avidin-HRPtechniques.Recommendedprimarydilutionsare1/1,000-1/2,000inPBS/0.3%TritonX-100-biotin/avidin-HRP.ByWesternblotanalysisofbrainhomogenatestheantibodyspecificallylabelsabandofapproximately155kD.Immunolabelingiscompletelyabolishedbypre-adsorptionwithsynthetichumannNOS(134-148)at5µgpermLofdilutedantibody.NocrossreactivitywithotherformsofNOSwasobserved.PhotoDescription:IHCimageofneuronsstainingfortheN-terminalofnNOSintheratcortex(aboveandbelowleft)andtheratstriatum(belowright).Thetissuewasfixedwith4%formaldehydeinphosphatebuffer,beforebeingremovedandpreparedforvibratomesectioning.FloatingsectionswereincubatedatRTin10%goatseruminPBS,beforestandardIHCprocedure.Primaryantibodywasincubatedat1:2000for48hours,goatanti-rabbitsecondarywassubsequentlyaddedfor1hourafterwashingwithPBS.Lightmicroscopystainingwasachievedwithstandardbiotin-streptavidin/HRPprocedureandDABchromogen,bottomleftimagewithnickelstain.
