BackgroundChemical Properties质量控制BackgroundCapecitabine（Xeloda）是一种抗癌化疗药物。Capecitabine (N4-pentyloxycarbonyl-5\'-deoxy-5-fluorocytidine)是一种新的氟嘧啶氨基甲酸酯，其通过位于肝脏和肿瘤中的三种酶转化为5-氟尿嘧啶（5-FUra）[1]。体外实验：在抗增殖实验中，当在与HepG2肝癌相同的板中培养LS174T WT和LS174T-c2细胞时，细胞对capecitabine更敏感。在LS174T WT单独和与HepG2培养时，capecitabine的IC50值分别为890 ± 48和630 ± 14 μM。当用与肝癌细胞相同的平板中培养LS174T-C2亚系时，IC50从330 ± 4下降到89 ± 6 μM。在LS174T-c2亚克隆中，capecitabine几乎不引起细胞死亡，早期和晚期凋亡分别增加了244和262％[1]。此外，Capecitabine以Fas依赖性方式诱导细胞凋亡，在胸腺嘧啶磷酸化酶（TP）转染的LS174T-c2细胞中，Capecitabine显示7倍的细胞毒性和显著更强的凋亡潜力[2]。在体实验：在CXF280、HCT116、COLO205和WiDr人结肠癌异种移植模型中，Capecitabine在更广的剂量范围内发挥功效[2]。在高度转移的裸鼠模型中，Capecitabine抑制肝癌切除后的肿瘤生长和转移性复发，这种结果归因于肿瘤中PD-ECGF的高表达[3]。参考文献：[1] Ishikawa T, Utoh M, Sawada N, et al.
