A collection of 7 agonists and modulators for a range of mGlu receptor subtypes. Products are freeze-dried as an exact weight so that addition of 1 ml or 0.5 ml of water provides a stock solution of 5 mM or 10 mM*, respectively.
The kit contains (S)-3,5-DHPG (ab120007), (R,S)-CHPG sodium salt (ab120221), LY 379268 (ab120196), L-AP4 (ab120002), VU0155041 sodium salt (ab120248), AMN082 dihydrochloride (ab120011) and (S)-3,4-DCPG (ab120006). *Please note that AMN082 dihydrochloride (ab120011) is only soluble to 5 mM in water.

(S)-3,5-DHPG:

Selective group I mGlu agonist.

mGlu5

(R,S)-CHPG sodium salt:

Water soluble form of (R,S)-CHPG (ab120039), a selective mGlu5 agonist.

Suggested Solvent: Water

mGlu2,3

LY379268:

Highly potent and systemically active mGlu2 and mGlu3 agonist. EC50 values are 2.69 and 4.48 nM for hmGlu2 and hmGlu3, respectively and displays 80-fold selectivity over group I and group III receptors. Effective in several animal models of stroke, epilepsy, drug abuse, schizophrenia, and pain.

L-AP4:

Selective group III mGlu receptor agonist.

mGlu4

VU0155041 sodium salt:

Water soluble, positive allosteric selective modulator at mGluR4 (EC50 values are 798 nM and 693 nM at human and rat mGluR4 receptors, respectively). It has advantages over PHCCC (ab120043) as it is water soluble and more potent. Active in behavioural models of Parkinsons Disease.

mGlu7

AMN082 dihydrochloride:

Potent, selective, orally active and brain penetrant mGlu7 receptor agonist. Potently inhibits cAMP accumulation and stimulates GTP binding (EC50 = 64-290 nM) in mGluR7 transfected cells. Selective over mGlu1, 2, 3, 4, 5, 6 and 8 and ionotropic receptors (up to 10 M). Elevates plasma stress hormones in vivo. Induces mGlu7 receptor internalisation in dissociated hippocampal neurones.

Suggested Solvent: Water (5 mM) / DMSO (10 mM)

mGlu8

(S)-3,4-DCPG:

Potent, selective mGlu8a agonist (EC50 = 31 nM). Displays 100-fold selectivity over mGlu7. Systemically active in vivo.

Providing storage is as stated on the product vial and the vial is kept tightly sealed, the product can be stored for up to 6 months.

Wherever possible, you should prepare and use solutions on the same day. However, if you need to make up stock solutions in advance, we recommend that you store the solution as aliquots in tightly sealed vials at -20 C. Generally, these will be useable for up to one week. Before use, and prior to opening the vial we recommend that you allow your product to equilibrate to room temperature for at least 1 hour.
