Capacity:forupto100testsOverviewMACSPlexCytokineAssaysaredesignedfordeterminingconcentrationsofupto7solubleanalytesinasinglesample.TheanalysisisbasedonMACSPlexCaptureBeads,whichdisplaydefinedfluorescencepropertiesandcanbeidentifiedusingstandardflowcytometrytechniques.DetailsTheMACSPlexCytokineBasicKitcontainsthesupplementarycomponentsnecessarytoperformaMACSPlexCytokineassay.IthastobecombinedwiththeMACSPlexCytokine12Standard,humanandthedesiredMACSPlexCytokineReagentsKits,humanorwiththeMACSPlexCytokine10Standard,mouseandthedesiredMACSPlexCytokineReagentsKits,mouse,respectively.Donotmixhumanandmousespecificitiesinoneexperiment.MACSPlexCytokineReagentsKitsaredesignedfordetectionofoneormaximaluptosevenanalytesofchoiceinasinglesample.MACSPlexCaptureBeadswithinthesingleMACSPlexCytokineReagentKitscontainfluorescentlylabeledbeadpopulations,coatedwithaspecificantibodyreactingwithoneoftheanalyteswithinthesample.MACSPlexCaptureBeadsfromdifferentreagentskitsdisplaydifferentfluorescenceintensities.Samplescontainingunknownlevelsofanalytesareincubatedwiththeantibody-coatedMACSPlexCaptureBeads,andanalytesbindtothespecificantibody.EachdetectionreagentcontainsAPC-conjugatedantibodiesspecificforoneanalyteandisalsoadded.Consequently,sandwichcomplexesareformedbetweentheMACSPlexCaptureBead,theanalyte,andthedetectionreagent.ThesecomplexescanbeanalyzedbasedonthefluorescencecharacteristicsofboththeMACSPlexCaptureBeadandthedetectionreagent.StandardsofknownquantitiesofgivenanalytesareprovidedwiththeMACSPlexCytokineStandardsandareusedforthequantificationoftheanalyteswithintheunknownsamples.TheanalysisofMACSPlexCustomCytokineAssaysrequireaflowcytometerwithablue(e.g.488nm)andared(e.g.635nm)laser,whicharecapableofdetectingFITC,PE,andAPC.IncombinationwiththeExpressModesoftheMACSQuant®AnalyzerstheMACSPlexCustomCytokineAssaysareoptimizedforautomatedmeasurementofthebeads.Theysimplifyflowcytometricanalysisviapredefinedexperimentsettingsaswellasacquisitionandanalysistemplates.Theyapplyanautomatedgatingstrategytopopulationsofinterestthatwillbeautomaticallyadjustedforeachdatafileindividuallytoachieveoptimalresults.ApplicationsMACSPlexCytokineReagentKits,humanhavebeendevelopedfortheflowcytometricdetectionofthesolublehumancytokinesGM-CSF,IFN-α,IFN-γ,IL-2,IL-4,IL-5,IL-6,IL-9,IL-10,IL-12p70,IL-17A,orTNF-αinasinglesample.Thekithasbeenoptimizedforusewithserum,plasma,andcellculturesupernatants.MACSPlexCytokineReagentKits,mousehavebeendevelopedfortheflowcytometricdetectionofthesolublemousecytokinesGM-CSF,IFN-γ,IL-2,IL-4,IL-5,IL-10,IL-12p70,IL-17A,IL-23,orTNF-αinasinglesample.Thekithasbeenoptimizedforusewithserum,plasma,andcellculturesupernatants.
