Exosomesaresmallmembranevesiclessecretedbymostcelltypes,andappearubiquitouslyincellculturesupernatantsandbodyfluids.Increasingevidencesupportsthatexosomesplayimportantrolesinintercellularcommunication,bothlocallyandsystemically,bytransportingtheircontentssuchasproteins,lipidsandRNAsbetweencells.Ofparticularinterestforcontrolleddrugdeliveryisthatcell-derivedexosomesofferthepossibilitiesofovercomingBIOLOGicalbarriers,therebyallowingtheincorporatedgeneanddrugtoreachtargetedtissue,whichhavebeenconsiderablechallengesforsyntheticcarriers.Greatresearcheffortshavebeendedicatedtodevelopingexosome-baseddrugdeliverysystemsforthetreatmentofinflammatorydiseases,degenerativedisordersandcancer.Inthisreview,wewilldescribethestructuralandfunctionalpropertiesofexosomesandemphasizecurrentadvancesinthetherapeuticapplicationsofexosomesasdrugdeliveryvehicles,followedbyadiscussiononcurrentchallengesandfutureperspectives.
