Background:Thereactivityoftheantiserumisrestrictedtolysozyme.InimmunoelectrophoresisandrADIalimmunodiffusion(Ouchterlony),usingvariousantiserumconcentrationsagainstnormalhumanmilkandtearsasingleprecipitinlineisobtainedwhichshowsareactionofidentitywiththeprecipitinlineobtainedwithpurifiedlysozyme.Noreactionisobtainedwithanyotherplasmaproteincomponentorserum.Inimmunocytochemicalandimmunohistochemicaltechniquesforthedetectionoflysozymeatthecellularandsubcellularlevelinappropriatelytreatedcellandtissuesubstrates.Thisimmunoconjugateisnotpre-diluted.Theoptimumworkingdilutionofeachconjugateshouldbeestablishedbytitrationbeforebeingused.Excesslabelledantibodymustbeavoidedbecauseitmaycausehighunspecificbackgroundstainingandinterferewiththespecificsignal.Source:Highlypurifiedrecombinanthumanlysozyme.Lysozyme,abacteriolyticenzyme,hasamolecularweightofabout15,000.Itsconcentrationinplasmaislow;inmilktheconcentrationisreportedtobebetween5and220mg/ml.Alsotearscontainaconsiderableamountoflysozyme.Freund"scompleteadjuvantisusedinthefirststepoftheimmunizationprocedure.Product:Biotin-coupledpurifiedhyperimmuneIgGlyophilizedfromasolutioninphosphatebufferedsaline(PBS,pH7.2)Nopreservativeadded,asitmayinterferewiththeantibodyactivity.Itisreconstitutedbyadding1mlsteriledistilledwater,spundowntoremoveinsolubleparticles,dividedintosmallaliquots,frozenandstoredatorbelow-20°C.Applications:ELISA,Immunocytochemistry,Immunohistochemistry(frozen),(In)directimmunofluorescence.CrossReactivity:Theantiserumdoesnotcross-reactwithanyothercomponentofhumanplasma.Inter-speciescross-reactivityisanormalfeatureofantibodiestoplasmaproteinssincetheyfrequentlyshareantigenicdeterminants.Cross-reactivityofthisantiserumhasnotbeentestedindetail.Specificity:Biotin-conjugatedIgGfractionofpolyclonalRabbitantiSerumtoHumanlysozyme.Storage:Thelyophilizedconjugateisshippedatambienttemperatureandmaybestoredat+4°C;prolongedstorageatorbelow-20°C.Priortouse,analiquotisthawedslowlyatambienttemperature,spundownagainandusedtoprepareworkingdilutionsbyaddingsterilephosphatebufferedsaline(PBS,pH7.2).Repeatedthawingandfreezingshouldbeavoided.Caution:ThisproductisintendedFORRESEARCHUSEONLY,andFORTESTSINVITRO,notforuseindiagnosticortherapeuticproceduresinvolvinghumansoranimals.Thisdatasheetisasaccurateasreasonablyachievable,butNordic-MubioacceptsnoliABIlityforanyinaccuraciesoromissionsinthisinformation.
