DescriptionCultrex®StemCellQualifiedHumanFibronectin,Pathclear®ispurifiedfromhumanplasmaandisPathclear®certifiedtobefreeofcommonpathogens.Fibronectinisanextracellularmatrixproteinthatisfoundabundantlyinbloodandconnectivetissues.Thesematricesareassociatedwiththeepithelialtomesenchymaltransitionofmetastaticcells,includingtumorcellswithstemcell-likeproperties.Fibronectinperformsessentialfunctionsincollagenfibrillogenesis,aseitherageneralcelladhesionmoleculeoramodulatorinbindingbetweencellsurfacesandtheextracellularmatrix.Fibronectinmatrixassemblyisessentialfornormalvertebratedevelopmentandapparentlycontributestothegenerationoftumormetastasesbysupportingtheestablishmentandpersistenceofpre-metastaticniches.Fibronectinissecretedasadisulfide-linkeddimerof230-270kDa,comprisedofthreetypesofrepeatingmodulesthatmediateinteractionswithextracellularmatrixcomponents(includingfibronectinitself),andcellsviaintegrinsandotherfibronectinreceptors.Thus,fibronectincanbeusedforcoatingtissueculturesurfacesorasamediumadditivetopromotecelladhesionandproliferation;itis0.2μmsterilefiltered.Cultrex®StemCellQualifiedHumanFibronectin,Pathclear®isapurifiedplasmaextracellularmatrix proteinthathasbeendeveloped,producedandqualifiedspecifically toprovideafunctionallydefinedandeffectivefeeder-freesurfacefortheattachmentandmaintenanceofembryonicstemcellsinapluripotentstateQualitycontrolspecifications:PathClear®–testednegativeforeighthumanpathogenicvirusesincludingHepatitisA,BandC,HIV1and2,Hantaan,Seoul,andSinNombrebyPCR.Nobacterialorfungalgrowthdetectedafterincubationat37°Cfor14daysfollowingUSPsterilitytestingguidelines.Endotoxinconcentration≤20EU/mlbyLimulusAmoebocyteLysate(LAL)assay.Proteinconcentrationof1mg/ml.NomycoplasmacontaminationdetectedbyPCR.Providedin100mMCAPS,150mMNaCl,1mMCaCI2,pH11.5.Purity ≥90%(SDS-PAGE).BIOLOGicalactivitydeterminedforeachlot:PromotestheattachmentofhumaniPSCs.EffectivelymaintainshumaniPSCsinapluripotentstateinafeeder-freeculture.SafetyStatement:AlthoughCultrex®StemCellQualifiedHumanFibronectin,PathClear®hasbeentestedforthevirusesaboveitcontainshumansourcematerialandthereforeshouldbetreatedaspotentiallyinfectiousandhandledattheBiologicalSafetyLevel2tominimizeexposure.
