InordertodeveloparapidandstraightforwardcouplingprocedureatthePEGterminus,amethodofdirectcouplingantibodiestothePEGterminusofliposomeswasintroducedbyBendasetal.Inthismethodology,antibodiesaresimplyattachedtothePEGterminusofliposomes,whichhadbeenendgroup-functionalizedwithcyanuricchloride,inmildbasicconditions(pH8.8)withoutpriorantibodyderivatizations.Ithasbeenshownthatinordertoobtainastableattachmentofproteinsonliposome,theDSPE-PEG-cyanurwasaddedintotheliposomestochemicallyconjugatewithproteinstoformastablecomplexandminimizethedenaturationofproteins.Proteinscanbecovalentlycoupledtotheliposomesviaamine-reactivecyanur-groups,eitherdirectlytothevesiclesurfaceusingcyanuricchloride-activatedDSPE(cyanur-DSPE)ortothedistalendsofPEG-spacersusingactivatedcyanur-PEG-PE(ammoniumsalt).CyanuricchlorideatthePEGterminusfunctionstolinkpeptides,antibodiesandotheramine-containingbiomoleculesornanoparticlesviaanucleophilicsubstitutionreactionunderbasicconditions.Antibodiesorotherproteinscanbeconjugatedwithoutanypreviousderivatization.Reactionbetweenliposomescontainingcyanurwithaminegrouponligand.ImmunoFluor™-CyanurisaPEGylatedproduct.Forotherreactive(PEGylatedandnon-PEGyalatedproducts)ImmunoFluor™productssuitableforothertypesconjugationmethodsseehere.
