DescriptionLRRK2isamemberoftheleucine-richrepeatkinasefamily.Itsroleisyetunknownbutitmayplayaroleinthephoshorylationofproteinscentraltoparkinsondiseases.LRRK2containsanankryinrepeatregion,aleucine-richrepeat(LRR)domain,akinasedomain,aDFG-likemotif,aRASdomain,aGTPasedomain,aMLK-likedomainandaWD40domain.LRRK2ispresentinthecytoplasmbutalsoassociateswiththemitochondrialoutermembrane.DefectsinLRRK2arethecauseofParkinsondisease8(PARK8).Parkinsondiseaseischaracterisedbybradykinesia,restingtremor,muscularrigidityandposturalinstABIlity,aswellasbyaclinicallysignificantresponsetotreatmentwithlevodopa.ThepathologyinvolvesthelossofdopaminergicneuronsinthesubstantianigraandthepresenceofLewybodies(intraneuronalaccumulationsofaggregatedproteins),insurvivingneuronsinvariousareasofthebrain.PARK8isanautosomal-dominantlate-onsetparkinsonism,characterizedbyonsetfrom50to65years,withslowprogressionandrelativelybenigncourse.RelatedproductsLeucine-RichRepeatKinase2inhibitorLRRK2-IN-1BatchNo.SeeproductlabelUnitsize50µgAntigenAsyntheticpeptide(CLKRKRKILSSDDSLRSS)correspondingtoaminoacids946-962ofthehumanLRRK2proteinconjugatedtodiptheriatoxinwasusedastheimmunogen.OtherNamesLeucine-richrepeatserine/threonine-proteinkinase2;Dardarin;LRRK2;PARK8AccessionQ5S007_HUMANProducedinRabbitPurityAffinitypurifiedApplicationsIHC.Adilutionrangeof1:500to1:1000isrecommended.Otherapplicationshavenotyetbeentested.Biosensisrecommendsoptimaldilutions/concentrationsshouldbedeterminedbytheenduser.SpecificitySpecificitywasdemonstratedbyimmunohistochemistry.TheantibodystainspositivetanglesininferiortemporalcortexofhumanbrainaffectedbyAlzheimer"sdisease.Cross-reactivityThisantiserumhasbeensuccessfullytestedinhuman.Otherspecieshavenotyetbeentested.BlastitIfyouwouldliketousethisproductinanotherspeciesotherthanthosespecifiedhere,ortoseethesharedidentitybetweentheimmunogenusedhereindifferentspeciesand/orothermolecules,simplycopytheimmunogen(fromtheImmunogenfield)andpasteitHEREandblast/formatit.Notethat,antiseraraisedagainstsyntheticpeptidesarequiteoftenveryspecificforthatpeptideie,onlyonesingleaminoaciddifferencemaybeenoughtorestrictthespecificitytoaparticularmolecule.FormLyophilisedReconstitutionReconstitutewith50µlsterilewaterandcentrifugetoremoveanyinsolublematerial.StorageAfterreconstitutionkeepaliquotsat-20ºCforahigherstability,andat4ºCwithanappropriateantibacterialagent.Glycerol(1:1)maybeaddedforanadditionalstability.Avoidrepetitivefreeze/thawcycles.ExpiryDate12monthsafterpurchaseReferences1.ZimprichA,etal.Neuron44:601-607(2004).2.OtaT,etal.Nat.Genet.36:40-45(2004).
